چکیده
Abstract Background Retinal degeneration is often accompanied by microglia-mediated neuroinflammation. Ferulic acid (FA), an active ingredient of traditional Chinese medicines (TCMs), has been reported to have anti-inflammatory effects. This study explores the impact FA on neuroinflammation and associated retinal in rd10 mice. Methods Rd10 mice received different concentrations every day from postnatal (P)4 P24. On P25, visual function was evaluated electroretinogram, retinae were collected for further investigation. Microglial activation expression relevant cytokines retina qPCR, western blotting immunofluorescence staining. structure assessed haematoxylin eosin (HE) Results Supplementation with 50 mg/kg provided optimal protection against degeneration, treated exhibiting more photoreceptor nuclei as well greater wave amplitude amplification electroretinogram than untreated suppressed microglial both vivo vitro, inhibited pro-inflammatory factors Tnf?, IL1?, Ccl2 Furthermore, STAT1 subsequently IRF8 expression, potentially highlighting a role these pathways FA-mediated immunomodulatory activity. Conclusions Attenuation may be beneficial retarding degeneration.

Contribution of Microglia-Mediated Neuroinflammation to Retinal Degenerative Diseases
Retinal degenerative diseases are major causes of vision loss and blindness worldwide and are characterized by chronic and progressive neuronal loss. One common feature of retinal degenerative diseases and brain neurodegenerative diseases is chronic neuroinflammation. There is growing evidence that retinal microglia, as in the brain, become activated in the course of retinal degenerative diseas...
متن کاملSpinal injection of docosahexaenoic acid attenuates carrageenan-induced inflammatory pain through inhibition of microglia-mediated neuroinflammation in the spinal cord.
Neuroinflammation in the spinal cord plays a critical role in the processing of inflammatory pain. Docosahexaenoic acid (DHA), a predominant omega-3 polyunsaturated fatty acid in the central nervous system, is known to modulate inflammatory responses in various neurodegenerative disorders. In this study, we investigated whether DHA could reduce inflammatory pain and inhibit neuroinflammation in...
متن کاملTetrahydroxystilbene Glucoside Attenuates Neuroinflammation through the Inhibition of Microglia Activation
Neuroinflammation is closely implicated in the pathogenesis of neurological diseases. The hallmark of neuroinflammation is the microglia activation. Upon activation, microglia are capable of producing various proinflammatory factors and the accumulation of these factors contribute to the neuronal damage. Therefore, inhibition of microglia-mediated neuroinflammation might hold potential therapy ...
متن کاملActivation of microglia and chemokines in light-induced retinal degeneration.
PURPOSE Microglial cells, which are activated and recruited by chemokines, have been shown to play crucial roles in neuronal degenerations of the central nervous system (CNS). This study investigated the activation and migration of retinal microglial cells and expression of chemokines in retinas in light-induced photoreceptor degeneration in mice. METHODS Ninety-five Balb/cJ mice were kept in...
متن کاملSafranal, a Saffron Constituent, Attenuates Retinal Degeneration in P23H Rats
Saffron, an extract from Crocus sativus, has been largely used in traditional medicine for its antiapoptotic and anticarcinogenic properties. In this work, we investigate the effects of safranal, a component of saffron stigmas, in attenuating retinal degeneration in the P23H rat model of autosomal dominant retinitis pigmentosa. We demonstrate that administration of safranal to homozygous P23H l...
